Abstract
We investigated the antioxidant properties of KR 31378 and its major metabolite, KR 31612, compared to ¥á-tocopherol and trolox. KR 31378 as well as ¥á-tocopherol strongly inhibited the production of thiobarbituric acid reactive substance (TBARS) induced by A7r5 cells with low density lipoprotein (LDL, 100 §¶/mg protein) and 0.1 mM CuSO4. The electron paramagnetic resonance (EPR) signals showed that KR 31378 and KR 31612 strongly and concentration-dependently scavenged both superoxide and hydroxyl radical. However, ¥á-tocopherol and trolox did suppress the EPR signals of hydroxyl radical but not superoxide. Based on the rate constants calculated from the slope of the linear regression plot, KR 31378 and KR 31612 exerted higher effectiveness than ¥á-tocopherol and trolox in scavenging hydroxyl radicals. In conclusion, these results suggest that KR 31378 was an effective scavenger of ROS and its radical scavenging property may be of clinical interest for vascular protection from oxidative injury as ¥á-tocopherol.
